I don't think it's a matter of knowing a certain tool. It's more a case of the applicant's general habits.

I don't use MS Word if I can avoid it. In fact, if a job that advertises that the writer will be working in MS Word, it becomes much less attractive to me. All the same, I routinely use styles the few times that I do use the program. As much as anything else, it's a habit that's ingrained very deeply because of my professional work.

While I wouldn't judge an applicant solely on the basis of whether they used styles or not in an MS Word resume, I can certainly see the point of taking that into consideration. After all, if people claim to be designers, then many people would judge them partly by the layouts of their resumes. So why shouldn't writers be judged on their work habits while writing? The use of styles speaks volumes about a person's knowledge or habits.
