If those were the only options, I'd choose Access and I'd probably read a book. However my own experience took a different path.

I learned Excel inside out and backwards, because I was writing books about it. In doing so, I also learned about the flat file database aspects of Excel and realized that Excel simply offered a different way to enter database data. That made recognizing what was what in Access a breeze. The jump to relational databases took place because I had to learn Oracle's, and again the grounding in Excel helped a great deal.

These days, I think it's important to learn the differences between a flat file database and a relational database, but I doubt you really need to get as up close and personal with either Excel or Access to do so. A good tutorial-level book would be a must, but so would access to a working copy of a relational database - few things beat hands-on experience.
