I think you can still download RoboHelp from their website and it is
free for 30 days. It gives you a chance to play around with it and
become more familiar with it. I am not sure about FrameMaker, but I will
make a leap and say that there is most likely a free download of the
software somewhere.

Yes, use samples of documentation you created from the past in your
portfolio.

As for skill sets, I have had the luxury of not having to learn many
platforms that are discussed on this board. In my three TW jobs--past
and current-- I was responsible for creating mostly internal
documentation, and a vast majority was hard copy. But some of the
<platforms> I have used are HTML, PDF, and the entire MS Office Suite,
mainly Word and Publisher.

I do have working knowledge of DOS, Linux, Unix, XML, and whatnot. I
have to use some of these when testing procedures I have written. But a
programmer I am not.

When I was in college, I did <play> with many of the TW tools you
mentioned and that are discussed on this list, but for the most part, I
personally have yet to use most of them outside the classroom. Again, I
have had three unique jobs, and not knowing these tools as much as the
next TW may prove to hinder me in the future, but that remains to be
seen. I would say, learn all you can.
