Here's my own experience:
There was an Ad in the Times of India for a Technical
Writer. The skill sets required fitted me to the T. So
confident was I of getting that job, that I sent the
cover letter with only one line it
"This job is mine. Thank you."
I didnot even get short-listed for an interview.
Lesson learnt: Not all risks pay-off.
