There's ALWAYS an opportunity to renegotiate. Whether it's fruitful or not
is the issue. In the OP's situation, I would have pushed for the original
rate that had been "promised," at which point the company may have replied
with the "take it or leave it" -- or they may have backed down and gone back
to the original rate, or counter-offered. So as you say, there's nothing to
lose by trying. If you don't make the attempt, then it is indeed a "take it
or leave it" by default.

There's certainly nothing to lose by trying, but my experience is that
if there was an opportunity to renegotiate you probably would have
gotten a call or email from someone telling you that they needed to
discuss their original rate with you. Changing the rate in the letter
with no advance notice was probably the same as "take it or leave
it," and I would be willing to bet you lunch that they had another
contractor ready to shop the job to if you had passed on it.
