A recruiter told me to ask at the end of the interview "Is there anything you can think of that would prevent me from moving forward in this position?" This accomplishes two things rather nicely:
- it indicates a serious interest in the position and
- identifies potential stumbling blocks *while there is still time to do something about them*

If the answer is no, the next question is "So what's the next step?" Inquiring, open, but not too pushy. If the answer is "Well, actually, we were looking for a candidate with experience with <toolname>," then you have a chance to counter with "I don't have that experience, but I learn tools very quickly and I can be up to speed before next week." Or whatever.
