I wouldn't refuse to respond to anything directed at me, but would keep the
responses short and to the point:
"Did you open my file?"
"No."
You opened my file, didn't you?"
"No."
etc.
OTOH, when the problem coworker stands in her cube and loudly shouts,
"Someone's opened my file! Who opened my file?" at the entire room, that's
the one that should go unanswered. After all, if you didn't open the file
she's not even talking to you, right?

Then, as Gene stated, if you are the file fiddler, then you should respond;
otherwise, if you are not fiddling with "her" files, there is no need for
you to say anything.
