I tried one for a while, and had mixed experience. I'm quite tall and found I had to use a 75cm ball to get high enough to activate my posture correctly - your thighs should make a 15 degree or so angle to the horizontal. You can deflate the balls to get a lower position, but remember that they also get much more bouncy when deflated. For comfort I preferred a harder, more inflated ball.
At first it makes you somewhat seasick as you tend to bounce a lot more, but you get used to that. In summer, sitting on a ball is uncomfortably hot and sweaty since they are made of a non-breathable material. Best to put a towel on the ball and sit on the towel. My main problem was that I couldn't adjust my desk high enough to get a sound ergonomic position (I couldn't get close enough under my desk) so I ended up abandoning the ball. If your desk is fully height adjustable then it would probably be a winner. Mind you, what I really wanted to try (and was impossible at my work) was a desk you could stand at.
